Role of odorant binding protein C12 in the response of Tribolium castaneum to chemical agents

Tribolium castaneum is a worldwide pest of stored grain that mainly damages flour, and not only causes serious loss of flour quality but also leads to deterioration of flour quality. Chemical detection plays a key role in insect behavior, and the role of odorant-binding proteins (OBPs) in insect chemical detection has been widely studied. OBPs can interact with small molecule compounds and thereby modulate variation in insecticide susceptibility in insects. In this study, a total of 65 small molecule compounds are selected to investigate the bound effect with TcOBP C12. The molecular docking results showed that β-caryophyllene, (−)-catechin, butylated hydroxytoluene, diphenyl phthalate and quercetin were the top five compounds, with docking binding energies of −6.11, −5.25, −5.09, −5.05, and − 5.03 Kcal/mol, respectively. Molecular dynamics analysis indicated that odorant binding protein C12 (TcOBP C12) exhibited high binding affinity to all five tested chemical ligands, evidenced by fluorescence quenching assay in vitro. In addition, the contact toxicity assay results suggested that these chemical agents caused a dose-dependent increase in mortality rate for T. castaneum adults. The TcOBP C12 gene was upregulated >2 times after a 24-h exposure, indicating that OBP C12 may play an important role for T. castaneum in response to these chemical agents. In conclusion, our results provide a theoretical basis for future insecticide experiments and pest management.

Odorant binding protein 18 increases the pathogen resistance of the imported willow leaf beetle, Plagiodera versicolora.

Insect odorant-binding proteins (OBPs) are a class of small molecular weight soluble proteins. In the past few years, OBPs had been found to work as carriers of ligands and play a crucial role in olfaction and various other physiological processes, like immunity. A subset of insect OBPs had been found to be expressed differently and play a function in immunity of fungal infection. However, there are few studies on the role of OBPs in immunity of bacterial infection. To identify the immune-related OBPs of Plagiodera versicolora after infected by Pseudomonas aeruginosa, we determined the mortality of P. versicolora to P. aeruginosa and selected the time point of 50% mortality of larvae to collect samples for RNA-seq. RNAi technology was used to investigate the function of immune-related OBPs after P. aeruginosa infection. RNA-seq data shows that PverOBP18 gene significantly up-regulated by 1.8-fold and further RT-qPCR affirmed its expression. Developmental expression profile showed that the expression of PverOBP18 was highest in the pupae, followed by the female adults, and lower in the 1st-3rd larvae and male adults with lowest in eggs. Tissue expression profiling showed that PverOBP18 was dominantly expressed in the epidermis. RNAi knockdown of PverOBP18 significantly reduced the expression of bacterial recognition receptor gene PGRP and antibacterial peptide gene Attacin and reduced the resistance of P. versicolora to P. aeruginosa infection. Our results indicated that PverOBP18 gene increased the pathogen resistance of P. versicolora by cooperating with the immune genes and provided valuable insights into using OBPs as targets to design novel strategies for management of P. versicolora.

Odorant binding protein C12 is involved in the defense against eugenol in Tribolium castaneum

Tribolium castaneum (T. castaneum) is a worldwide pest of stored grain that mainly harms flour, and not only causes serious loss of flour quality but also leads to deterioration of flour quality. Chemical detection plays a key role in insect behavior, and the role of odorant-binding proteins (OBPs) in insect chemical detection has been widely studied. However, the mechanism of OBPs in insect defense against exogenous toxic substances is still unclear. In this study, biochemical analysis showed that eugenol, the active component of A. vulgaris essential oil, significantly induced the expression of the OBP gene OBPC12 from T. castaneum (TcOBPC12). The mortality of late larvae treated with eugenol was higher than that of the control group after RNA interference (RNAi) against TcOBPC12, which indicates that the OBP gene is involved in the eugenol defense mechanism and leads to a decrease in sensitivity to eugenol. Tissue expression profiling showed that the expression of TcOBPC12 in the epidermis, hemolymph, and intestine was higher than in other larval tissues, and TcOBPC12 was expressed mainly in the epidermis, head, and fat body of adults. The developmental expression profile showed that the expression of TcOBPC12 in late eggs, early and late larval stages, and late adult stages was higher than in other developmental stages. These data suggest that TcOBPC12 may be involved in the absorption of exogenous toxic substances by the larvae from T. castaneum. Our results provide a theoretical basis for the metabolism and degradation mechanism of exogenous toxic substances and help explore more potential target genes of insect pests.

Combined transcriptomic, proteomic and genomic analysis identifies reproductive-related proteins and potential modulators of female behaviors in Spodoptera litura

The common cutworm, Spodoptera litura, is a polyandrous moth with high reproductive ability. Sexual reproduction is a unique strategy for survival and reproduction of population in this species. However, to date available information about its reproductive genes is rare. Here, we combined transcriptomics, genomics and proteomics approaches to characterize reproductive-related proteins in S. litura. Illumina sequencing in parallel with the reference genome led to the yields of 12,161 reproductive genes, representing 47.83% of genes annotated in the genome. Further, 524 genes of 19 specific gene families annotated in the genome were detected in reproductive tissues of both sexes, some of which exhibited sex-biased and/or tissue-enriched expression. Of these, manual efforts together with the transcriptome analyses re-annotated 54 odorant binding proteins (OBPs) and 23 chemosensory proteins (CSPs) with an increase of 18 OBPs and one CSP compared to those previously annotated in the genome. Interestingly, at least 35 OBPs and 22 CSPs were transcribed in at least one reproductive tissue, suggestive of their involvement in reproduction. Further proteomic analysis revealed 2381 common proteins between virgin and mated female reproductive systems, 79 of which were differentially expressed. More importantly, 74 proteins exclusive to mated females were identified as transferred relatives, coupled with their specific or high expression in male reproductive systems. Of the transferred proteins, several conserved protein classes across insects were observed including OBPs, serpins, trypsins and juvenile hormone-binding proteins. Our current study has extensively surveyed reproductive genes in S. litura with an emphasis on the roles of OBPs and CSPs in reproduction, and identifies potentially transferred proteins serving as modulators of female post-mating behaviors.

The Role of the Peripheral Environment of Neuronal Receptors in Stimuli Perception by Insects’ Sense Organs: Facts and Hypotheses

The role of intermediate substrates represented by morphological structures or chemical compounds located between the information carrier (stimulus) and the dendrite receptor membrane of insect sense organs is considered an example of olfactory, visual, mechanical, hygro-, and thermoreceptors. Intermediate substrates in olfactory sensillae are represented by their cuticular regions, pores or pore-tubular system, sensillum lymph, and pheromone-binding proteins. Intermediate structures also imply articular membrane (mechanoreceptor hairs), tympanic membrane (hearing organs), mineral statoliths (gravity receptors), iron oxide nanoparticles (magnetic field receptors), matrix surrounding the dendrites (hygroreceptors), microparticles associated with the dendrite membrane (thermoreceptors), and nonsclerotized mesocuticle (infrared receptors). There are two stages in propagation of a signal that is perceived by sense organs of most modalities: (1) before a signal contacts the peripheral environment (substance or structure) and (2) after a signal contacts the peripheral environment. Besides, a signal of one modality on the first stage of its propagation can be replaced by a signal of another modality at the second stage of propagation, as, for example, in hygro- or thermoreceptors, since the primary stimulus (moisture, heat/cold, or infrared radiation) is replaced by a mechanical effect on the dendrite membrane of its peripheral environment. The mechanisms of signal modality substitution in many sense organs, as well as the role of odorant-binding proteins and pore tubules in olfactory sensillae, have not been fully elucidated and require further investigation.

Odorant-Binding Proteins Contribute to the Defense of the Red Flour Beetle, Tribolium castaneum, Against Essential Oil of Artemisia vulgaris.

The function of odorant-binding proteins (OBPs) in insect chemodetection has been extensively studied. However, the role of OBPs in the defense of insects against exogenous toxic substances remains elusive. The red flour beetle, Tribolium castaneum, a major pest of stored grains, causes serious economic losses for the agricultural grain and food processing industries. Here, biochemical analysis showed that essential oil (EO) from Artemisia vulgaris, a traditional Chinese medicine, has a strong contact killing effect against larvae of the red flour beetle. Furthermore, one OBP gene, TcOBPC11, was significantly induced after exposure to EO. RNA interference (RNAi) against TcOBPC11 led to higher mortality compared with the controls after EO treatment, suggesting that this OBP gene is associated with defense of the beetle against EO and leads to a decrease in sensitivity to the EO. Tissue expression profiling showed that expression of TcOBPC11 was higher in the fat body, Malpighian tubule, and hemolymph than in other larval tissues, and was mainly expressed in epidermis, fat body, and antennae from the early adult. The developmental expression profile revealed that expression of TcOBPC11 was higher in late larval stages and adult stages than in other developmental stages. These data indicate that TcOBPC11 may be involved in sequestration of exogenous toxicants in the larvae of T. castaneum. Our results provide a theoretical basis for the degradation mechanism of exogenous toxicants and identify potential novel targets for controlling the beetle.

Mating-based regulation and ligand binding of an odorant-binding protein support the inverse sexual communication of the greater wax moth, Galleria mellonella (Lepidoptera: Pyralidae).

In moths, sex pheromones play a key role in mate finding. These chemicals are transported in the antennae by odorant-binding proteins (OBPs). Commonly, males encounter conspecific females; therefore, several OBPs are male-biased. Less is known, however, about how the olfactory system of moths has evolved toward inverse sexual communication, ie where females seek males. Therefore, the objective of this study was to identify the profile of OBPs and their expression patterns in the bee hive pest, Galleria mellonella, a moth that uses inverse sexual communication. Here, OBP-related transcripts were identified by an RNA Sequencing (RNA-Seq) approach and analysed through both Reverse Transcription Polymerase Chain Reaction (RT-PCR) in different tissues and quantitative real-time PCR for two states, virgin and postmating. Our results indicate that G. mellonella has 20 OBPs distributed amongst different tissues. Interestingly, 17 of the 20 OBPs were significantly down-regulated after mating in females, whereas only OBP7 was up-regulated. By contrast, 18 OBP transcripts were up-regulated in males after mating. Additionally, binding assays and structural simulations showed general odorant-binding protein 2 (GOBP2) was able to bind sex pheromone components and analogues. These findings suggest a possible role of OBPs, especially GOBPs, in the inverse sexual communication of G. mellonella, with gene expression regulated as a response to mating.

Silencing of Odorant-Binding Protein Gene OBP3 Using RNA Interference Reduced Virus Transmission of Tomato Chlorosis Virus.

Tomato chlorosis virus (ToCV) is widespread, seriously impacting tomato production throughout the world. ToCV is semi-persistently transmitted by Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae). Currently, insect olfaction is being studied to develop novel pest control technologies to effectively control B. tabaci and whitefly-borne virus diseases. Despite current research efforts, no report has been published on the role of odorant-binding proteins (OBPs) in insect preference under the influence of plant virus. Our previous research showed that viruliferous B. tabaci preferred healthy plants at 48 h after virus acquisition. In this study, we determined the effect of OBPs on the host preference interactions of ToCV and whiteflies. Our results show that with the increase in acquisition time, the OBP gene expressions changed differently, and the OBP3 gene expression showed a trend of first rising and then falling, and reached the maximum at 48 h. These results indicate that OBP3 may participate in the host preference of viruliferous whiteflies to healthy plants. When the expression of the OBP3 gene was knocked down by an RNA interference (RNAi) technique, viruliferous Mediterranean (MED) showed no preference and the ToCV transmission rate was reduced by 83.3%. We conclude that OBP3 is involved in the detection of plant volatiles by viruliferous MED. Our results provide a theoretical basis and technical support for clarifying the transmission mechanism of ToCV by B. tabaci and could provide new avenues for controlling this plant virus and its vectors.

Characterization and expression profiling of odorant-binding proteins in Anoplophora glabripennis Motsch

In insects, olfaction plays a critical role in locating hosts, recognizing mates, and selecting oviposition sites. The Asian long-horned beetle (Anoplophora glabripennis Motschulsky) feeds on 43 species of trees in 15 families, but its chemosensory mechanisms are poorly understood. Herein, genes encoding 61 odorant-binding proteins (OBPs) were identified from the published genome and our previous A. glabripennis transcriptomic data. To investigate their physiological functions, we performed expression profiling of all AglaOBPs in the antennae, legs, and maxillary palps of both sexes. Phylogenetic analysis clustered A. glabripennis OBPs into four subgroups, comprising 29 Minus-C OBPs, 15 Antennae-binding proteins (ABPIIs), 10 Classic OBPs, and one Plus-C OBP. 12 AglaOBP genes were expressed specifically in antennae, and AglaOBP3, AglaOBP18, AglaOBP21, AglaOBP33, AglaOBP41, AglaOBP45, and AglaOBP47 were particularly highly expressed in male antennae. These proteins may function in the detection of female sex pheromones. AglaOBP23 and AglaOBP44 were preferentially expressed in maxillary palps. Expression profiling suggests that many OBPs may be involved in olfaction and gustation, in addition to carrying hydrophobic molecules. The AglaOBPs family has acquired functional diversity concurrently with functional constraints, and further investigation could provide insight into the roles of OBPs in chemoreception.

Identification and Expression Profile Analysis of Odorant Binding Proteins in the Oriental Fruit Fly Bactrocera dorsalis

Olfaction is crucial in many insects for critical behaviors, including those regulating survival and reproduction. Insect odorant-binding proteins (OBPs) function in the first step of the olfactory system and play an essential role in the perception of odorants, such as pheromones and host chemicals. The oriental fruit fly, Bactrocera dorsalis, is a destructive fruit-eating pest, due to its wide host range of up to 250 different types of fruits and vegetables, and this fly causes severe economic damage to the fruit and vegetable industry. However, OBP genes have not been largely identified in B. dorsalis. Based on our previously constructed B. dorsalis cDNA library, ten OBP genes were identified in B. dorsalis for the first time. A phylogenetic tree was generated to show the relationships among the 10 OBPs of B. dorsalis to OBP sequences of two other Dipteran species, including Drosophila melanogaster and the mosquito Anopheles gambiae. The expression profiles of the ten OBPs in different tissues (heads, thoraxes, abdomens, legs, wings, male antennae and female antenna) of the mated adults were analyzed by real-time PCR. The results showed that nine of them are highly expressed in the antenna of both sexes, except BdorOBP7. Four OBPs (BdorOBP1, BdorOBP4, BdorOBP8, and BdorOBP10) are also enriched in the abdomen, and BdorOBP7 is specifically expressed in leg, indicating that it may function in other biological processes. This work will provide insight into the roles of OBPs in chemoreception and help develop new pest-control strategies.

Comparative Genomics of Odorant Binding Proteins in Anopheles gambiae, Aedes aegypti, and Culex quinquefasciatus

About 1 million people in the world die each year from diseases spread by mosquitoes, and understanding the mechanism of host identification by the mosquitoes through olfaction is at stake. The role of odorant binding proteins (OBPs) in the primary molecular events of olfaction in mosquitoes is becoming an important focus of biological research in this area. Here, we present a comprehensive comparative genomics study of OBPs in the three disease-transmitting mosquito species Anopheles gambiae, Aedes aegypti, and Culex quinquefasciatus starting with the identification of 110 new OBPs in these three genomes. We have characterized their genomic distribution and orthologous and phylogenetic relationships. The diversity and expansion observed with respect to the Aedes and Culex genomes suggests that the OBP gene family acquired functional diversity concurrently with functional constraints posed on these two species. Sequences with unique features have been characterized such as the “two-domain OBPs” (previously known as Atypical OBPs) and “MinusC OBPs” in mosquito genomes. The extensive comparative genomics featured in this work hence provides useful primary insights into the role of OBPs in the molecular adaptations of mosquito olfactory system and could provide more clues for the identification of potential targets for insect repellants and attractants.

The Odorant Binding Protein Gene Family from the Genome of Silkworm, Bombyx mori

BackgroundChemosensory systems play key roles in the survival and reproductive success of insects. Insect chemoreception is mediated by two large and diverse gene superfamilies, chemoreceptors and odorant binding proteins (OBPs). OBPs are believed to transport hydrophobic odorants from the environment to the olfactory receptors.ResultsWe identified a family of OBP-like genes in the silkworm genome and characterized their expression using oligonucleotide microarrays. A total of forty-four OBP genes were annotated, a number comparable to the 57 OBPs known from Anopheles gambiae and 51 from Drosophila melanogaster. As seen in other fully sequenced insect genomes, most silkworm OBP genes are present in large clusters. We defined six subfamilies of OBPs, each of which shows lineage-specific expansion and diversification. EST data and OBP expression profiles from multiple larvae tissues of day three fifth instars demonstrated that many OBPs are expressed in chemosensory-specific tissues although some OBPs are expressed ubiquitously and others exclusively in non-chemosensory tissues. Some atypical OBPs are expressed throughout development. These results reveal that, although many OBPs are chemosensory-specific, others may have more general physiological roles.ConclusionSilkworms possess a number of OBPs genes similar to other insects. Their expression profiles suggest that many OBPs may be involved in olfaction and gustation as well as general carriers of hydrophobic molecules. The expansion of OBP gene subfamilies and sequence divergence indicate that the silkworm OBP family acquired functional diversity concurrently with functional constraints. Further investigation of the OBPs of the silkworm could give insights in the roles of OBPs in chemoreception.

Role of Odorant Binding Proteins: Comparing Hypothetical Mechanisms with Experimental Data

Odorant binding proteins are expressed in the olfactory epithelium of many mammalian species and are thought to assist in the uptake and transfer of odorants from the gas phase to the odor receptors. Various mechanisms have been proposed to explain how mass transfer occurs. Most experimental work has focused on the binding of ligands to odorant binding proteins or to the olfactory receptors under steady-state conditions, whereas the situation in vivo is dynamic due to the tidal flow of air through the nose and the mass transfer in and out of the olfactory epithelium. Some preliminary dynamic data have been obtained from an in vitro system, and this has been used, along with other published data, to test some of the proposed mechanisms for odorant binding protein (OBP). An assessment of the hypothetical mechanisms has been made to examine how well the mechanisms fit with the published experimental data. A new hypothesis is proposed where OBP acts to prolong the odor signal and increases the signal output of the olfactory system.
